Route 45 is a state highway in the U.S. state of New Jersey. It runs 28.51 miles (45.88 km) from Route 49 in Salem to U.S. Route 130 in Westville. It was originally Route 6 which was created in 1916, and in the 1927 renumbering, it became Route 45.

Route 45 begins in Salem, at Route 49. It passes by the Memorial Hospital of Salem County just prior to becoming concurrent with County Route 540. The concurrency lasts for over a mile before splitting up, and proceeding northward as it intersects County Route 653. It intersects County Route 669, and just yards later, County Route 631. It then intersects County Route 601, County Route 668 and County Route 617, just before intersecting County Route 616.[2][1]

Route 45 travels near the Salem County Veterans Cemetery shortly before reaching a concurrency with US 40 as it enters the Boro of Woodstown. It soon intersects County Route 657 before intersecting County Route 660, while turning just east of north. Route 45 then intersects County Route 694. Route 45 then intersects County Route 607 shortly before meeting a junction with County Route 617. It intersects County Route 538, before it becomes concurrent with Route 77, shortly before US 322 also becomes concurrent with Route 45. Around 0.5 miles later, US 322 splits from Route 45 as it enters Woodbury. It intersects County Route 664, before intersecting County Route 664, County Route 603 and County Route 634.[2][1]

Route 45 then intersects County Route 627, County Route 678, County 632 and Alt County Route 553. It then intersects County Route 648 and County Route 656, before intersecting the New Jersey Turnpike. Shortly after, it intersects County Route 650, County Route 551 and County Route 663. Soon, it intersects County Route 644, County Route 642 and County Route 553. It then intersects Interstate 295, shortly before splitting up into two parts, and then ending at US 130.[1][3]

Today's Route 45 was originally Route 6, which was created in 1916. In the 1927 renumbering, Route 6 between Camden and Salem was renumbered as Route 45. In 1929, a bypass around Camden was created as the southern half of Crescent Boulevard. In the 1953 renumbering, Route 45 remained.[4]

In the late 1960s, the Delaware Valley Regional Planning Commission (DVRPC) proposed construction of a 10-mile-long, $20 million freeway along the Route 45 corridor in Gloucester County which would have extended from Exit 24 of Interstate 295 in Woodbury, to be completed by 1985. The New Jersey Department of Transportation never followed through on this proposal, instead proposing a widening project to create a four-lane divided highway for the full length of Route 45 from Woodbury to Salem, of which only a small portion was ever completed.[5]
