There are 40 songs in the main Career mode for the PlayStation 2 version, and 48 songs in the Xbox 360 version. Songs are organized by tiers that are related to the song's difficulty, and the arrangement is different for both the PlayStation 2 and the Xbox 360 version. Progression at a selected difficulty mode for Guitar Hero II has changed from the original Guitar Hero. Each tier must be completed in order to unlock the next tier. To complete a tier, the player must complete a fixed number of songs depending on difficulty level. When the player successfully completes the last required song, they are offered an option to play the encore song for that tier. The player can decline this, which still unlocks the encore song, but the next tier is not accessible until the encore is completed.
Once a song is unlocked in a tier at any difficulty, it is available for the quick play, competitive, and co-operative modes.
Most songs in the game are covers of the original songs, but there are some master recordings as indicated below. Each song has both a lead guitar track and a bass guitar track; some songs also possess a rhythm guitar track, as indicated below.
Songs not present in the PlayStation 2 game are indicated by "--" in the PS2 Tier column.

There are 25 bonus songs in the PlayStation 2 version of Guitar Hero II and 2 additional songs for the Xbox 360 version. All bonus songs are unlockable by using in-game money to buy them within the game's store. Once purchased, they are playable in quick play, competitive, and co-operative modes. The song "Raw Dog" by The Last Vegas was selected as part of the "Be a Guitar Hero" content for the release of Guitar Hero II. All bonus songs are master recordings, and where marked, include rhythm guitar in addition to the lead and bass guitar tracks.

The Xbox 360 version of the game allowed from the creation of downloadable content available through Xbox Live Marketplace, typically in packs of three songs.[2] Some of the songs include those that were included in Guitar Hero, and thus are covers of those songs, while the other packs were new material and consisted of master recordings. In some cases, a free video preview was available separately.

- Note 1: While the original "You Really Got Me" was performed by the band The Kinks, the information from Activision specifically lists the cover by Van Halen for inclusion in the game.
- Note 2: Pack sizes and cost are for all songs within the pack.
- Note 3: The name of this download track contains a mistake. It is listed as "Exile - by Soilwork from Germany" on Xbox Live Marketplace[8] and on the RedOctane corporate website RedOctaneGames.com[2]. However, the band is actually from Sweden.[10] Soilwork's record label, Nuclear Blast Records, has a subsidiary in Germany.[11]
