From its southern terminus, County Route 507 runs northward along Schuyler Avenue. For several blocks, the west side of the road is in Harrison, while the east side is in Kearny; soon, the entire road is in Kearny. After a little over two miles, the highway reaches the Belleville Turnpike, NJ 7, and it turns left to run westward concurrently with NJ 7, along the border with Bergen County. Along this one-mile concurrency is the southern terminus of NJ 17.

One mile west of Schuyler Avenue, CR 507 leaves NJ 7 and turns northward onto River Road, following the Passaic River. River Road continues northward through North Arlington and into Lyndhurst, where it becomes Riverside Avenue. After passing through Rutherford, the highway crosses the old right-of-way of the Erie Railroad at Carlton Hill and enters East Rutherford, becoming Carlton Avenue. CR 507 then turns right onto Locust Lane and heads into Wallington, where it becomes Locust Avenue and then joins Main Street.

At a three-way intersection, CR 507 bears left onto Midland Avenue, where it crosses the Saddle River and enters Garfield. It shortly turns left onto River Drive and continues north, following the Passaic River. In Elmwood Park, the road passes beneath the Garden State Parkway and has interchanges with US 46, I-80, and NJ 4. In Fair Lawn, the highway becomes known as River Road.

Near the north end of Fair Lawn, CR 507 leaves the Passaic River and turns right onto Maple Avenue. The road continues through Glen Rock and Ridgewood, and then crosses the Hohokus Brook into the borough of Ho-Ho-Kus.

At the first traffic light in Ho-Ho-Kus, CR 507 turns left to pick up the Franklin Turnpike and continues north, following an old alignment of NJ 17. For a short distance in Ho-Ho-Kus and Waldwick, CR 507 runs concurrently with CR 502. CR 507 continues north through Allendale, Ramsey, and Mahwah, crossing NJ 17 in Ramsey. Just before the New York state line, US 202 merges into the Franklin Turnpike, which after about 200 feet becomes Orange Avenue in Suffern, New York.
