Maryland Route 16 is a two-lane road running initially north-south connecting Caroline County, Maryland, with neighboring Dorchester County, Maryland, but veering into an east-west pattern after crossing the county line south.

After meeting Delaware Route 16 near Hickman, the road travels in a northwesterly direction, joining with Maryland Route 404 for several miles before turning sharply south near Williston. The road passes through Preston, joining Maryland Route 318 and heading east for a few miles before again veering south into Dorchester County. It then merges into U.S. Route 50 outside of East New Market before turning left at Cambridge. Ten miles after Cambridge, Route 16 ends in Taylors Island at Hoopers Neck Road. In total, Route 16 is 51 miles long.

Sites of note that the road serves include the Linchester Mill (circa 1682), a veteran's cemetery in Beulah, the home of Revolutionary War hero William Richardson, and the ancestral farm of Charles Dickinson, infamous for his duel with Andrew Jackson.
