Samantha grew up mainly in the foster care system in one of Boston's roughest neighborhoods after her parents died within months of each other. Despite her hard life, she was able to graduate high school with excellent marks and receive a scholarship to Charles University, thanks to her benefactor, Dorothy O'Sullivan.

Upon reaching Charles University, Samantha Best thought her college life would be an enlightened and different experience from her troubled childhood. However, on her first day she had meal plan troubles, clashes with her roommate and involvement in an accidental death. Kathryn Klarner, her roommate who instigated the accidental death of Sam's love interest Devon Sylver's roommate John, had been angry with Sam after being spurned by her. For revenge, Kathryn spread a rumor that Sam ratted out the parties involved in the rooftop death. After this, Sam became something of a social pariah.
Eventually, Sam went to the dean to apologize for her wrong-doing. The dean, Ms. Dymond, was ready to expel Sam, until Ms. O'Sullivan stepped in and told Ms. Dymond that if she expelled Sam, she would take back her generous donation to the university. It was around this time Sam befriended Dawn Vargaz who helped her find work at local hot spot Colony, which solved her financial issues. Also, around this time she befriends Noah Jensen, the archetypal film nerd, and Cynthia, a shy girl she neglected originally who was secretly molested by her father, a professor at Charles University.
She also happens to be adopted, breaks up with her boyfriend Devon, because he is extremely jealous, and quits her job at Colony. After breaking up with Devon she learns more about Trent, including his cocaine addiction,(Colony was shutdown as a result of Trent's drug connections with drug dealers who were using Colony to sell drugs)and at first is disappointed at him, but resolves to help him through his problems, because someone has to. Despite his defects Trent as proven to be real supportive of Sam and helps her find her birth parents. In the episode, Mommy Dearest, she goes to her birth mother's house and mets her for the first time. When the family of Sam's mom comes home, she tells Sam to leave and that it was a mistake to let her met her. This causes Sam to become sad and Trent comforts her by saying she is not a mistake, and the two have sex in one of showers in the dorm bathrooms. When Trent shows signs of unable to handle the relationship, Sam decides to let him go for a month to think things over, and tells Trent that once she comes back he has to prove to her that he does want a relationship with her, and she drives away with Dawn.
