Pennsylvania Route 75 is a 76 mile long north-south state highway located in central Pennsylvania, USA. The southern terminus is at the Mason-Dixon Line in Montgomery Township, where PA 75 continues into Maryland as Maryland Route 494. The northern terminus is at an interchange with U.S. Route 22 and U.S. Route 322 in Port Royal.

From 1927 to 1928, PA 75 was signed on the current alignment of Pennsylvania Route 28 from Brookville to Brockway.
