Route 27 is a highway in extreme northeastern Missouri. Its northern terminus is the Iowa state line over the Des Moines River where the road continues as Iowa Highway 27 (for which it was numbered after). Its southern terminus is at U.S. Route 61 west of Alexandria. This route was formerly Route B and U.S. Route 136 Spur. Wayland is the only town on the highway.

This route is also part of the Avenue of the Saints that stretches from St. Paul, Minnesota, to St. Louis, Missouri.
