Route 27 is a north-south Massachusetts (United States) state route that runs from Kingston to Chelmsford. The route serves some of Boston's southern, western, and northwestern suburbs.
Route 27 is directly served by interchanges from state routes 24, 9, and 2. I-90 and I-95 only serve the route indirectly via interchanges with nearby roadways.

Route 27's original form was identical to the modern route south of Wayland, but north of there it followed current Route 126 into Concord. It came into its current form by 1939.
