M-49 is a short north-south route in the south-central portion of the U.S. state of Michigan that goes from Litchfield south to the Ohio state line. M-49 is the only Michigan highway in which the road crossing into another state has the same state highway number. M-49 continues as SR 49, which is the westernmost interchange with the Ohio Turnpike. SR-49 begins at US 35 in Dayton, Ohio and runs up the west side of the state, connecting with M-49.

M-49 passes through Allen where it crosses US 12 (the old US 112). It also passes through Reading, and Camden. Near the Michigan/Ohio state line just south of Camden, it has a junction with Territorial Road, a Hillsdale county road that used to be M-120. Territorial Road becomes SR 120 as it crosses the Indiana state line in the short segment of north-south border between Indiana and Michigan.[3]

M-49 was added to the state highway system in 1920, running along its current routing between Litchfield and the Ohio state line. The northern terminus at the time was located on M-9 which is now M-99. The now defunct M-120 ran concurrently with M-49 from 1939 until 1946. This concurrency was removed when M-49 was realigned to smooth out some curves in the road.[2]
