State Road 550 exists in two portions. The western section is completely within Knox County and has a western terminus east of Emison at U.S. Highway 41 and runs southeasterly to the U.S. Highway 50/U.S. Highway 150 concurrency in Wheatland. State Road 241 terminates at its northern end on the opposite side of US 50/150 at the same intersection.
The eastern section terminates at both ends at US 150 in Martin County between Loogootee and Lacy.
